Inhibition of Translation by Methylglyoxal

چکیده

Methylglyoxal was a weak inhibitor of translation in the r e t i c u l o c y t e l y s a t e c e l l f r e e system and it did not display cap-dependent inhibition. A similar inhibition was obtained in a wheat-germ cei l f ree system that d i sp layed extensive cap-dependent inhibition with the cap ana logue 7 m e t h y J g u a n o s i n e phosphate . These results show that the chemical reaction of methylglyoxat with 7-methylguanosine is not the mechanism for the inhibition of protein synthesis by methylglyoxal and that methylglyoxat is a weak general inhibitor of translation.

Selective inhibition of mitochondrial respiration and glycolysis in human leukaemic leucocytes by methylglyoxal.

The effect of methylglyoxal on the oxygen consumption of mitochondria of both normal and leukaemic leucocytes was tested by using different respiratory substrates and complex specific artificial electron donors and inhibitors. The results indicate that methylglyoxal strongly inhibits mitochondrial respiration in leukaemic leucocytes, whereas, at a much higher concentration, methylglyoxal fails ...

Inhibition of electron flow through complex I of the mitochondrial respiratory chain of Ehrlich ascites carcinoma cells by methylglyoxal.

The effect of methylglyoxal on the oxygen consumption of Ehrlich-ascites-carcinoma (EAC)-cell mitochondria was tested by using different respiratory substrates, electron donors at different segments of the mitochondrial respiratory chain and site-specific inhibitors to identify the specific respiratory complex which might be involved in the inhibitory effect of methylglyoxal on the oxygen consu...

Inhibition of succinic dehydrogenase by methylglyoxal.

The reaction of the carbonyl group of methylglyoxal with -SH radicals of thiols has been the subject of numerous investigations (l-9). The reactivity of the keto aldehyde with -SH compounds suggested that a similar type of reaction may occur between methylglyoxal and -SH proteins. The physiological r&e of methylglyoxal as an inhibitor of -SK enzymes is of particular interest, since this keto al...

Methylglyoxal in cells elicits a negative feedback loop entailing transglutaminase 2 and glyoxalase 1☆

Glyoxalase 1 (GlxI) is the key enzyme that converts the highly reactive α-oxo-aldehydes into the corresponding α-hydroxy acids using l-glutathione as a cofactor. In our preliminary data, GlxI was identified as a substrate of transglutaminase 2 (TG2), a ubiquitous enzyme with multiple functions. According to the catalytic properties of TG2, protein cross-linking, polyamine conjugation, and/or de...

Increased dicarbonyl metabolism in endothelial cells in hyperglycemia induces anoikis and impairs angiogenesis by RGD and GFOGER motif modification.

Chronic vascular disease in diabetes is associated with disruption of extracellular matrix (ECM) interactions with adherent endothelial cells, compromising cell survival and impairing vasculature structure. Loss of functional contact with integrins activates anoikis and impairs angiogenesis. The metabolic dysfunction underlying this vascular damage and disruption is unclear. Here, we show that ...
